چکیده
Abstract Hepatocellular carcinoma (HCC) has a desmoplastic and hypoxic tumor microenvironment (TME), resulting in poor prognosis treatment resistance. This study aimed to construct novel prognostic classifier investigate the degree of hypoxia immune profiles HCC. Patients with HCC from public databases were classified into three HIF-1α clusters according 16 reported HIF-1α-related genes. Then, an score system was constructed based on nine overlapping differentially expressed genes (ODEGs) among various clusters. cluster. Besides, oncologic pathways infiltration also investigated HCCs different scores. The reliable predictive abilities for patients’ survival impressively suggested by significant C-indexes ROC analysis. All enrolled tumors divided high-, medium-, low-HIF-1α groups. Compared other two groups, high group exhibited highest enrichment multiple oncogenic pathways, such as TNF-α signaling via NF-кB, IL6-JAK-STAT3 signaling, mTORC1 MYC Hedgehog signaling. Notably, higher scores correlated advanced immunosuppressive TME. represented non-response rate transcatheter arterial chemoembolization (TACE) checkpoint blockade (ICB). In conclusion, we developed distinguish profiles.

Metformin suppresses hypoxia-induced stabilization of HIF-1α through reprogramming of oxygen metabolism in hepatocellular carcinoma
Overexpression of hypoxia-induced factor 1α (HIF-1α) has been shown to be involved in the development and progression of hepatocellular carcinoma (HCC). HIF-1α should therefore be a promising molecular target for the development of anti-HCC agents. Metformin, an established antidiabetic drug, has proved to also be effective in treating cancer although the precise underlying mechanisms of this a...
متن کاملPrognostic Significance of HIF-1α Expression in Hepatocellular Carcinoma: A Meta-Analysis
AIM Pilot studies have evaluated the correlation between hypoxia-inducible factor-1α (HIF-1α) overexpression and clinical outcome in hepatocellular carcinoma (HCC) patients. However, the results remain inconclusive. To comprehensively and quantitatively summarize the evidence on the suitability of HIF-1α to predict the prognosis of patients with HCC, a meta-analysis was carried out. METHODS S...
متن کاملPrognostic value of DNA repair based stratification of hepatocellular carcinoma
Aberrant activation of DNA repair is frequently associated with tumor progression and response to therapy in hepatocellular carcinoma (HCC). Bioinformatics analyses of HCC data in the Cancer Genome Atlas (TCGA) were performed to define DNA repair based molecular classification that could predict the prognosis of patients with HCC. Furthermore, we tested its predictive performance in 120 indepen...
متن کاملHypoxia inducible factor 1α gene (HIF-1α) splice variants: potential prognostic biomarkers in breast cancer
BACKGROUND Hypoxia-inducible factor 1 (HIF-1) is a master transcriptional regulator of genes regulating oxygen homeostasis. The HIF-1 protein is composed of two HIF-1alpha and HIF-1beta/aryl hydrocarbon receptor nuclear translocator (ARNT) subunits. The prognostic relevance of HIF-1alpha protein overexpression has been shown in breast cancer. The impact of HIF-1alpha alternative splice variant ...
